Is there a way to reduce the overall colour saturation by 50% whilst performing a QTGMC de-interlace?

Sure, by using the "Filtering->Vapoursynth->Color->Tweak" with "Saturation" at "0.50". Whether QTGMC is used or not is independet of other filters. (In case you are using Avisynth: Tweak is also available in the Avisynth filter tab.)

The tweak setting seems to have vanished? Is it now somewhere else?

Still there under "Filtering->Vapoursynth->Color->Tweak" (assuming 'Filtering->Support' is set to Vapoursynth)
